496 Solar simulated light induces cutaneous squamous cell carcinoma in inbred mice: A clinically relevant model to investigate T cell responses

چکیده
Over one million cases of cutaneous squamous cell carcinoma (cSCC) are diagnosed annually in the United States, and approximately 4% patients develop metastases 2% die. Yet, there is a paucity clinically relevant cSCC murine models. Outbred SKH-1 mice highly susceptible to solar simulated light (SSL)-induced cSCC. However, an outbred strain limits ability evaluate MHC-restricted, antigen-specific T responses perform studies with genetically engineered mice. To address this need, inbred FVB/N, BALB/c, C57BL/6J were exposed 42 weeks SSL (cumulative dose: 856 kJ/m2 UVB 9,884 UVA). 22 257 2,967 reliably induced tumors FVB/N BALB/c median onset 38 41 weeks, respectively, delayed compared In contrast, only 15% developed apparent tumors. Most histologically as actinic keratosis, situ, or invasive Invasive had significantly decreased percentages tumor-infiltrating dendritic cells, CD4+ IFN-γ+ CD8+ granzyme B+ regulatory cells situ Progression from early late corresponded transition immune-activating immunosuppressive microenvironment. This SSL-induced model will facilitate future investigating anti-tumor genes influencing tumor formation response therapy.

[Solar elastosis in cutaneous squamous cell carcinoma].
INTRODUCTION Solar elastosis, or basophilic degeneration of collagen, may be a histologic sign of chronic sun damage. MATERIAL AND METHODS We reviewed 222 cases of squamous cell carcinoma (SCC) to identify the presence of solar elastosis and its possible invasion of the upper, middle, or deep reticular dermis. We also analyzed clinical variables such as SCC location, location in exposed areas...
متن کاملThe Usefulness of CD10 in Distinguishing between Cutaneous Basal Cell Carcinoma and Squamous Cell Carcinoma
Background & Objectives: CD10 is a cell surface enzyme with metalloendopeptidase activity, also known as Common Acute Lymphoblastic Leukaemia Antigen, which mainly serves as a marker for acute lymphoblastic leukemia (ALL). To date and to the best of our knowledge, only few comparative immunohistochemical studies have assessed CD10 expression in cutaneous epithelial neoplasms. Our goal was to d...
متن کاملCytodiagnosis of Cutaneous Basal and Squamous Cell Carcinoma
Background and Objective: Cutaneous cytology is the study of scraping or smears obtained from skin lesions. Although not commonly employed, cutaneous cytology has been a useful tool for clinicians due to simplified procedures and high sensitivity and specificity of presurgical clinical diagnosis of commo...
متن کاملTissue eosinophilia in oral and cutaneous squamous cell carcinoma and normal oral and cutaneous tissues
Introduction: Eosinophils produce proteases that stimulate invasion in cancers. Due to the more malignant potential of oral squamous cell carcinoma (OSCC) compared to cutaneous squamous cell carcinoma (CSCC), the aim of the present study was to compare tissue eosinophilia between OSCC and CSCC samples to understand whether tissue eosinophilia play a role in different biological...
متن کاملTongue Squamous Cell Carcinoma: A Clinical Study
Background and Objectives: Squamous cell carcinoma of the tongue (SCCOT) is the most common malignant neoplasm in oral cavity. At the time of the diagnosis of SCCOT regional metastases with a high mortality rate are common, which is due to the extensive lymphatic system drainage of the tongue. Although the level of healthcare has significantly improved, the survival rate is still low with an...
